In this experiment,field growthlicoricetender leavesa monthas materials,effects of various factors onseparationofliquoriceleaf protoplasts,these factors include theenzyme solutioncomponents,enzymolysis time,osmotic pressure anddissociation.Through the experiment discovered:young leaves ofGlycyrrhizasolutionenzymolysiseffect is the bestin 2% cellulase,pectinase0.5%,its yieldis 11.5 ×104 /g.FW;protoplastwasstandingby thedissociation of12hmost,amount to 11.6× 104 /g.FW;osmotic pressure of 0.35M enzyme solution on the separation in the best licorice leaf protoplast,protoplast yield was 11.1 × 104 /g.FW; with static and slowly oscillating enzyme obtained by protoplast the best quality, the highest yield, 12.5 × 104/g.FW. This study lays a foundation for protoplast culture andsomatic hybridization oflicorice.
